q APPLICATIONS

 

VivaMagic^® Transfection Reagent는 Non-liposomal transfection reagent로서 DNA와 complex를 이뤄 동물세포나 곤충세포에 transport할 수 있는 multi-component reagent 입니다.

 

일반적인 여러 가지 종류의 동물세포에 높은 transfection efficiency를 보여줍니다.: HeLa, NIH/3T3, COS-1, COS-7, CHO-K1, Hep G2 등.

 

특히 HEK-293 동물세포의 경우는 매우 우수한 transfection efficiency를 보여주며 transient 및 stable transfection에 적합합니다.

 

Serum 유무에 관계없이 우수한 transfection 효율을 보여주며 cytotoxic effect가 거의 없어 media를 바꾸어 줄 필요성이 없습니다.

 

q SHORT WORKING PROTOCOL:

 

60-mm dish 이용한 293T cell의 경우

1.     세포준비: 50~80 % confluency

2.     DNA준비: 0.5~2.0 ㎍

3.     Mixture 준비:Tube에 DNA와 serum-free medium넣어 94 ㎕로 맞추고 6 ㎕의 VivaMagic^® Transfection Reagent를 첨가하여 최종 100 ㎕로 맞춘다. (tapping 방법으로 mixing)

4.     Incubation: 상온에서 10~45 min 방치

5.     배양용기 medium 표면에 한 방울씩 첨가

6.     48시간 후 harvest & assay (cytotoxic effect가 거의 없어 harvest때까지 media 교체가 필요 없음)

q PRODUCT DESCRIPTION

 

VivaMagic^® Transfection Reagent는 non-liposomal reagent로서 동물세포유래 어떠한 성분도 포함되어 있지 않은 차세대 transfection reagent 입니다. 타겟 동물세포에 생리적 (physiological), 형태적 (morphological) 변화를 거의 주지 않고 동물세포막을 통한 transfer를 수행함으로써 효율적인 transfection을 보여줍니다. 또한 철저한 품질관리를 통해 안정적이고 재현성 있는 실험수행을 가능하게 합니다.

q STORAGE & STABILITY

 

VivaMagic^® Transfection Reagent의 보관온도는 2~8 ℃이며 배송 시 ice-pack 용기에 싸여 있습니다. 특히 0 Co이하 냉동상태 보관 시 transfection효율이 저하될 수 있으므로 유의하여 주시기 바랍니다. 보증기간은 1년입니다.

 

q SPECIAL HANDLING

 

- 사용 전 VivaMagic^® Transfection Reagent를 항시 잘 섞어 주시고 사용 전 상온에서 10분 이상 방치해 주십시오.

- VivaMagic^® Transfection Reagent를 다른 Vial에 aliquot하지 말아 주십시오.

- 플라스틱 Vial에 남아있는 약간의 chemical residue들은 본 시약의 biological activity를 저하시킬 수 있기 때문입니다.

- Serum-free medium과 섞을 때 반드시 medium과 직접 닿게끔 pipetting하시고 dilution step에서 serum-freemedium과 혼합 시 tube벽에 흘리지 않기 바랍니다.

 

* Note: 본 제품은 인체에 유해할 수 있으므로 피부접촉이나 흡입 등에 주의 하십시오.또한, 실험용 목적으로만 사용하여 주시고 진단용으로는 사용을 금해 주시기 바랍니다.

 

q EXPERIMENTAL PROCEDURE

 

- 본 실험 절차는 일반적인 transfection protocol과 유사합니다.

- 6-well plate 또는 35mm culture dish를 기준으로 작성되어 있습니다.

- 정확한 혼합을 위해 polystyrene계 tube를 사용하시고 tube내벽에 묻지 않도록 유의 하십시오.

Suspension cells (부유성 세포)	Transfection 당일 2ml media에 5 X 105 cells/ml~1 X 106 cells/ml 농도로 세포수를 맞추어 준비한다.

**DNA양 및 VivaMagicT^® Transfection reagent양은 조건에 따라 변화될 수 있고 DNA purity는 최소 OD 1.6이상을 권장합니다.
***60-mm dish 기준시 DNA 2 ug 당 VivaMagic^® Transfection reagent 6 ul를 사용 권장하며 DNA양은 실험목적에 따라 변경할 수 있으나 DNA: VivaMagic^® Transfection reagent의 ratio는 1:3(w/v)을 권장합니다.

 

1.     Transfection 할 세포준비
Transfection할 세포준비는 부착성세포 (adherent cell)과 부유성세포 (suspension cell)에 따라 다르기 때문에 효율적인 transfection을 위해 상기 Table 1을 참고로 준비한다. VivaMagic^® Transfection Reagent, DNA, diluent를 사용 전 상온에서 10분 이상 방치시키고 VivaMagic^® Transfection Reagent는 사용 전 1~2초간 부드럽게Tapping한다. 이때 diluent로는 serum free media를 사용하도록 한다.

2.     Transfection Complex (Mixture) 준비
항생제가 들어있지 않은 serum-free medium에 DNA 1~2㎍을 넣어 97 ㎕ volume이 되도록 맞추고VivaMagic^® Transfection Reagent 3.0 ㎕를 첨가하여 최종 100 ㎕가 되도록 맞춘다. Culture vessel에 따른Mixture준비는 Table 2를 참조한다.

3.     Mixture Incubation
준비된 mixture를 tapping하여 혼합한 다음 상온에서 10~40분간 반응시킨다.

* Note 1. 96-well plate 사용시 혼합한 후 5~10초간 shaker를 사용하여 rotating시킨 후 15분간 반응시킨다.

* Note 2. 다른 culture vessel의 경우 transfection complex준비는 상기 table 2를 참조한다.)

4.     Transfection complex 첨가
세포배양기에서 culture vessel을 꺼내고 배양배지를 제거하지 않은 상태에서transfection complex를 한 방울씩 medium 표면에 떨어뜨리면서 culture vessel을 부드럽게 swirling시킨다.
* Note 1. transfection complex를 배지에 넣고 난 후 cytotoxic effect가 거의 없기 때문에 media를 fresh 배지로 교체할 필요가 없음.
* Note 2. 96-well plate 경우 낮은 속도에서 30초간 shaking시킨다.
* Note 3. serum 포함 배지로 교체를 원할 경우 transfection 후 3~8시간 사이에 배지교체를 수행한다.

5.     Cell culture & Assay
Transfection 후 18~48시간 배양한 다음 assay를 수행한다.

 

q 주의사항

 

1.     Incubation time은 cell type, cell medium, cell density, 발현고자 하는 protein type에 따라 다를 수 있습니다.

2.     Stable transfection 실험을 위해서는 transfection 후 24 ~ 48시간에 계대 배양하고 다음날 목적에 맞는 항생제를 넣고 수일 ~ 수주간 선별 배양합니다.

3.     Transfection reagent: DNA의 optimal ratio와 transfection complex total volume은 cell line, cell density, assay등 조건에따라 다를 수 있으므로 조건확립을 위한 선행실험이 필요 할 수 있습니다.
